EPA Multiple-Path Particle Dosimetry Model (US EPA) release:

The EPA released the EPA MPPD v2.0 model on July 31st. This version of the multiple-path particle dosimetry (MPPD) model underwent external peer review and modernizes the derivation of inhalation dosimetry adjustment factors (DAF) applied in various assessment workflows to translate exposures to internal dose in different regions of the respiratory tract and refine dose-response analyses.  

 

It replaces the use of the Regional Deposited Dose Ratio (RDDR) model implemented by EPA in 1994 for most species. New features in EPA MPPD v2.0 include revised clearance rates for all regions of the respiratory tract in order to predict retained mass in humans, rats and mice. The model supports customization to various human use cases and provides an ability to enter solubility parameters if available. A revised approach for the estimation of systemic delivery is also provided. Use cases are illustrated and supported by template files to organize simulation data and Excel files to perform calculations. This new version is well positioned to serve as the basis for interoperability with other types of models such as computational fluid dynamics / computational fluid particle dynamics (CFPD) and PBPK to extend its prediction capabilities to inhaled gases, account for absorption, distribution, metabolism and elimination (ADME), or provide localized estimates to specific respiratory tract tissues. The website for download of the final document describing the model development and illustrative use cases, together with templates for recording and reporting simulations, Excel spreadsheets to perform calculations typically deployed in inhalation assessment workflows, and the link to obtain the model which was released on Friday (7/31) is at this URL:
